PLATELETS AND VASCULAR OCCLUSION Adenosine diphosphate as a mediator of platelet aggregation in vivo : an editorial view

چکیده
Platelets in hemostasis When blood vessels are injured so that they bleed, circulating platelets adhere to the damaged vessel walls and aggregate, so diminishing or arresting the hemorrhage. This interaction between platelets and vessel walls therefore has an easily demonstrable physiologic function. There is much clinical and experimental evidence that a deficiency or defect in circulating platelets is associated with "spontaneous" hemorrhages from small vessels. This suggests that platelets are somehow essential for the functional integrity of these vessels, but if there is such a mechanism it has yet to be established.

Multiple alterations of platelet functions dominated by increased secretion in mice lacking Cdc42 in platelets.
Platelet activation at sites of vascular injury is crucial for hemostasis, but it may also cause myocardial infarction or stroke. Cytoskeletal reorganization is essential for platelet activation and secretion. The small GTPase Cdc42 has been implicated as an important mediator of filopodia formation and exocytosis in various cell types, but its exact function in platelets is not established. He...

OBJECTIVE Adenosine signaling is known to inhibit platelet aggregation. Extracellular adenosine mainly stems from enzymatic phosphohydrolysis of precursor nucleotides via ecto-5'-nucleotidase. Previous studies suggest that soluble 5'-nucleotidase (5'-NT) derived from Crotalus atrox venom may be clinically beneficial in vascular leakage, myocardial, renal, and intestinal ischemia, or acute lung ...

Thrombosis, especially arterial, develops by mediation of platelets, adhering to collagen fibers at areas of endothelial cell damage. At this phase, platelet activation leads them to secrete factors [ADP, Fibrinogen, von Willebrand Factor (vWF), Fibronectin, Factor XIII] capable of promoting cell aggregation/adhesion and coagulation, inducing vasoconstriction (thromboxane A2) and acting as mito...
متن کاملAnti-Platelet Activities of Polyozellin In Vitro and In Vivo
Purpose: Thrombosis and thromboembolic occlusions of blood vessels are a major complication in various peripheral vascular diseases. The agents for the inhibition of platelet function are recognized as key tools in the treatment of atherothrombosis. Therefore, it became a mainstay medication for a wide range of vascular diseases. Polyozellin (POZ), a major constituent of an edible mushroom Poly...
متن کاملAn investigation of the antiplatelet effects of succinobucol (AGI-1067).
Succinobucol is a phenolic antioxidant with anti-inflammatory and antiplatelet effects. Given the importance of oxidant stress in modulating platelet-platelet and platelet-vessel wall interactions, the aim of this study was to establish if antioxidant activity was responsible for the antiplatelet activity of succinobucol. Platelet aggregation in response to collagen and adenosine diphosphate (A...
